A precise balance between excitatory and inhibitory synaptic activity is crucial for the proper function of the mammalian neocortex. Cortical inhibitory GABAergic interneurons are central for maintaining this balance by modulating network activity, and disturbances in their development have been linked to a range of neuropsychiatric disorders. Consequently, the identification and characterization of the molecular determinants governing their development is highly relevant for understanding corticogenesis under both physiological and pathological conditions. The present Suppl. Videos show different aspects and approaches for investigating this topic. Videos 1 and 2 display time-lapse footages of murine organotypic brain slices and the intracortical migration behavior of Sst-Cre/tdTomato interneurons compared to Sst-Cre/tdTomato/Dnmt1 loxP2 cells at the embryonic stage 14.5. Videos 3 and 4 display time-lapse material of single Cerebellar Granule Cells after control- or Dnmt1-siRNA treatment concerning their talin-GFP dynamics in vitro. Videos 5-11 show different epileptic events during PTZ experiments in adult mice. Video 12 shows a stereotypical behavior in an adult mouse indicative of an autism-like phenotype.
