We used Tandem Mass Tags (TMT) 11-plex coupled to 2D-LC-MS/MS to profile EDL muscles from WT and SOD1<sup>G37R </sup>mice at presymptomatic (P380) and symptomatic (P480) stages. This strategy not only achieved high specificity and sensitivity, evidenced by robust enrichment of NMJ proteins and ALS-linked signatures, but also establishes a powerful framework to capture synaptic vulnerability and disease progression at the molecular level.
