The microbial community of liquid cultures, containing wheat straw as a sole carbon source, were investigated over eight weeks using ITS and 16S sequencing

Two-liter shake flasks, containing 1 L minimal media and 5% (w/v) milled wheat straw, were inoculated with 1% (w/v) compost. The inoculum was collected from composting wheat straw that had been developed over the period of a year and watered at regular intervals. Genomic DNA was harvested from the compost cultures using a modified CTAB protocol adapted, and ITS and 16S regions were amplified before being sequenced on an ion torrent platform.
