To examine the potential off-target effects by CRISPR/Cas13d-mediated CTSL knockdown in 293FT cells Overall design: 293FT cells were transfected with non-targeting pre-gRNA control(gCtrl) or pre-gRNA targeting CTSL(gCTSL) expression plasmid in combination with Cas13d expression plasmid for 24h. RNA-seq was performed to characterize the differential expressed genes between gCtrl and gCTSL groups.
