To examine the role of the TGF-beta/Smad3 pathway in cardiac damage by doxorubicin, we performed comparitive RNA sequencing analysis of cardiac endothelial cells isolated from WT and Smad3 knockout mice. Experimental male mice utilized in the study received one or four weekly tail vein injections of doxorubicin, 5 mg/kg each, starting at 7 weeks of age. Control mice were injected with saline. Cardiac endothelial cells were isolated from mice treated with doxorubicin or saline 9 weeks after the last injection. RNA sequencing analysis was performed on the isolated cells.
