Shotgun sequencing of hankyphage capsids

In order to investigate the DNA packaging of the hankyphage, we performed PEG-precipitation of lysates of the B. thetaiotaomicron jmh43 strain and the jmh43::dcas9-repcHP strain (which exhibits and enhanced hankyphage induction). This was followed by a DNAse and Benzonase treatment and a subsequent SDS treatment to open the capsids and expose the DNA. Of note, jmh43::dcas9-repcHP lysate was sequenced using a PCR-free kit, whereas for jmh43 a previous step of PCR amplification was required to obtain enough DNA for Illumina sequencing.
