The HIF target MAFF promotes tumor invasion and metastasis through IL11 and STAT3 signaling

Using RNA-sequencing and ChIP-sequencing, we identified direct target genes of the transcription factor MAFF in MDA-MB-231 under normoxia and hypoxia. Overall design: MDA-MB-231, a human breast cancer cell line, was treated either under normoxia or hypoxia. After ChIP or RNA extraction, sequencing was performed. After knocking down endogenous MAFF in MDA-MB-231, we overexpressed V5-tagged MAFF with mutations on shRNA target sites. Then nuclei was extracted and we proceeded to ChIP procedures using V5 antibody to further determine direct targets of MAFF.
