Due to their ligand profiles, it can be hypothesized that Stab1 and Stab2 may be involved in atherosclerosis. We treated mouse aorta endothelial cells (MAOEC) with plasma from Stabilin-deficient and Wildtype mice to account for gene changes. We used microarrays to detail the global programme of gene expression in MAOEC during different plasma treatments. We identified distinct classes of up- and down-regulated genes in response to the treatment. Primary mouse aorta endothelial cells (MAOEC) (Pelo Biotech; cat. no. C57-6052) were grown to confluence in 6-well plates supplied with Endothelial Cell Medium (Pelo Biotech) containing 10% FCS. Subsequently, the Endothelial Cell Medium was supplemented with 10% plasma of ApoE-KO, ApoE-Stab1-KO and ApoE-Stab2-KO mice after 8 weeks of Western Diet treatment instead of FCS. After 48 hours of incubation, the RNA of the MAOEC was isolated.
