SINEUP Nanopore m6A. N6-methyladenosine profiling on SINEUP non-coding RNA by Nanopore Sequencing

We employed Nanopore targeted direct RNA sequencing in order to map m6A methylation sites on miniSINEUP RNA. Taking advantage of A549 cells stably transduced with inducible ShMETTL3 and ShCtrl lentiviral vectors, we compared the sequencing electric signals of transfected IVT miniSINEUP-DJ-1 RNA in the presence and absence of METTL3 expression. As a non-modified control, we also used the very same IVT miniSINEUP-DJ-1 RNA spiked in total RNA extract from A549 ShCtrl cells.
