The mantling abnormality is a somaclonal variation risk that is a major concern in oil palm clonal propagation. Based on previous cloning data, leaf tissues from selected oil palms (ortets) that had generated high mantling rate in their clones were sampled. Small RNA and 3'mRNA sequencing was carried out on these samples. Leaf samples from ortets that generated low mantling rates were included for comparison. As mantling was previously associated to the karma element in EgDEF1 gene, targeted bisulfite sequencing of the karma-EgDEF1 region was also carried out on these samples.
