To investigate the differential expressed circRNAs, and even their biological functions in human proximal tubular epithelial (HK-2) cells induced by normal glucose, high levels of glucose and treatment of dapagliflozin, metformin and vildagliptin. HK-2 cells were cultured in DMEM medium supplemented with 10% fetalfoetal bovine serum, and 100 U/mLU/ml penicillin, and 0.1 mg/mLl streptomycin at 37°C ℃ under humidified air with 5% CO2. After synchronized by culturing in DMEM for 24 h, the cells were treated with normal glucose (5.6mM), high glucose (30.0 mM) for 24 h, then treated with Dapagliflozin (2.5 μM), Metformin (5 mM), Vidagliptin (150 μM) for 48 h in 6-well plates. All groups were repeated three times.
