Ligand induced receptor activation of Tyro3, Axl and Mertk (TAM) leads to distinct signaling pathways that may favor oncogenesis via molecular various mechanisms. However, comparison of these three receptors for gene expression changes that is brought about by their activation alone has turned out be elusive mainly because of their distinct preferences to multiple ligands (GAS6, Protein S and Phosphatidylserine) and overlapping expressions in cell lines. To understand the role of ligand induced gene expression changes by TAM in a normalized ligand inducible manner, we constructed chimeric EGFR-TAM receptors that contained extracellular EGFR domains and intracellular TAM kinase domains. These receptors were stably and comparably expressed in EGFR-negative Chinese Hamster Ovarian cell lines. Post-serum starvation, cells were induced with equal amount of EGF for 6 and 24 hours, total RNA was extracted and subjected to RNASeq and further analysis for distinct or overlapping changes in gene expression by TAM kinases.
