De novo transcriptome sequencing and gene mining of polysaccharide metabolism in Lentinula edodes

Lentinan,is the most important polysaccharide isolated from Lentinula edodes. Previous studies mainly focused on the extraction, purification, characterization and function analysis of Lentinan. The metabolic pathways of polysaccharide and the key genes affecting the metabolism of polysaccharide in Lentinula edodes are still difficult to be resolved. In this research, we performed comparative de novo transcriptomic analysis among Lentinula edodes fruiting bodies at different growth stages by using high-throughput Illumina RNA-sequencing to analysis the molecular mechanisms underlying polysaccharide metabolism in Lentinula edodes. We found out 6 genes involved in the synthesis and degradation of polysaccharide by KEGG pathway analysis. Among these, gls and glg played a crucial role in polysaccharide metabolism of Lentinula edodes friuting bodies.
