We performed single cell sequencing using the Parse Biosciences WT kit to evaluate changes in dendritic cell populations following induction of experimental autoimmune encephalomyelitis. Mice in this experiment previously received CD8+ T cells from donor mice that had been immunized with PLP178-191 or OVA323-339. Thus we ask what the effect of these CD8s is on DC populations in the context of EAE. Dendritic cells were harvested from the spleens and inguinal lymph nodes of mice that received CD8+ T cells from mice immunized with either OVA323-339 or PLP178-191. DC harvest was performed 12 days following induction of experimental auotoimmune encephalomyelitis. Cells were fixed using the Cell Fixation Kit and processed with the Evercode Whole Transcriptome Mini Kit from Parse Biosciences. Sequencing was performed on the Illumina NovaSeq 6000 using 100bp paired-end reads.
