The Bradyrhizobium japonicum USDA110 transposon mutant library, randomly integrated by Tn5-derivative plasmid pLG107 (GenBank: KF532966.1), was generated to carry out the routine genotype-phenotype screening experiments. Several phenotypic transposon mutants failed to be complemented, and one possible explanation is the imprecise transposon localization information provided by PCR methods. The unbiased short reads library was thus constructed against each genome of transposon mutant strain and was submitted for illumina high-throughput re-sequencing. The short reads produced from each mutant strain were then applied to determine the transposon localization.
