Genome-wide MUSIC

We generated a genome-wide MUtational SIgnature Catalog by knocking out each gene in the mouse genome and obtaining a mutation signature on a defined CRISPR induced double-strand break. Approximately 90,000 barcodes targeting about 19,000 genes were tested on 3 different loci and 2 replicates. Candidate genes were chosen to create a subset which was retested, but now at increased resolution. In addition this project contains validation loci in mouse, mainly on HPRT as well as targeting human cell lines for candidate genes that were followed up.
