E. coli strain K12 was evolved in the presence of antimetabolite chemotherapies (5-FU or FUDR) in nutrient-poor or nutrient-rich media. Drug resistance was confirmed for each evolved strain. Whole genome sequencing was performed on four individually evolved E. coli strains from each drug/media condition: 5-FU/poor-media, 5-FU/rich-media, FUDR/poor-media, FUDR/rich-media, along with the ancestor strain. Additionally, RNA-sequencing was used to determine the transcriptional profiles of two 5-FU/nutrient-poor and two 5-FU/nutrient-rich evolved strains. mRNA was extracted from strains during exponential growth phase in their respective media without drugs.
