The research begins with the theoretical and instrumental foundations of molecular spectroscopy, highlighting how Raman and IR techniques provide distinct yet complementary molecular fingerprints. Moving beyond the direct method of microbes detection and structural identification, we investigates dynamic biochemical processes using a metabolically responsive dye system resazurin and its reduced form, resorufin as a marker for microbial activity. Through a multi-spectroscopic approach combining Raman, IR, fluorescence, and UV–Vis spectroscopy with Density Functional Theory (DFT) calculations. To further enhance detection limits and spatial precision, Surface-Enhanced Infrared Absorption (SEIRA) and SERS were integrated with OPTIR spectroscopy, enabling ultrasensitive detection of resorufin with submicron resolution. The power of OPTIR was then extended to plant biochemistry, where it enabled label-free, spatially resolved imaging of cyanide-containing metabolites in sorghum leaves.
