We identified 13 preferred terms (PTs) related to herpes virus infections and categorized them into all herpes events, HSV-related events, VZV-related events, and severe herpes events (eczema herpeticum, EH). For each category, we calculated four disproportionality metrics: reporting odds ratio (ROR), proportional reporting ratio (PRR), empirical bayes geometric mean (EBGM), and information component (IC). In addition, subgroup analyses by sex and age were applied. Time-to-onset (TTO) was calculated from treatment start to event onset. See Supplementary table 1-3 for details.
The sex and age subgroup analyses show that age and sex should be integrated into individual patient risk assessment and monitoring strategies. Females showed higher susceptibility to most herpetic infection–related events. Abrocitinib signals peaked in adolescents (0–17 years), while patterns in adults (18–64 years) resembled the overall population. In the elderly (≥65 years), baricitinib showed stronger signals for all herpes and VZV events (supplementary figure1-2)
Supplementary 1-3 presents the classification criteria for 13 PT terms, the specific numerical values and calculation methods for each major category under each drug, as well as the standards for the four positive signal indicators. Supplementary Figure 1-2 presents the results of the subgroup analysis by gender and age.
