The HSS cDNA sequences of Crassocephalum crepidioides accession Ile-Ife were obtained by (I) PCR using redundant primer pairs, (II) 3´- and 5´-RACE and (III) primer pairs allowing amplification of the cDNA. The obtained cDNAs were heterologously expressed in E. coli and enzymatic activity of the experessed homospermidine synthases confirmed by in vitro reactions using spermidine and putrescine as substrates. The formation of homospermidine was confirmed by HPLC analysis.
