Previously, we evaluated the relationship between neighborhood deprivation index (NDI)—a measure of neighborhood socioeconomic deprivation—and pro-inflammatory cytokines (e.g., IL-1β, IL-6, IL-8, TNFα, and IFNγ) and clinical markers (e.g., LDL, HDL) in a community-dwelling African American cohort at-risk for CVD from the Washington, D.C. Cardiovascular Health and Needs Assessment (DC-CHNA). We found significant associations between NDI and IL-1β and TNFα independent of body mass index (BMI) and atherosclerotic cardiovascular disease (ASCVD) 10-year risk score (Farmer et al., 2021). Notably, the relationship between NDI and HDL trended toward significance. To further investigate the potential relationship between neighborhood socioeconomic deprivation and HDL in the DC-CHNA cohort, we examined the relationships between NDI and multiple HDL characteristics (cholesterol, particle number, size, subspecies) from lipoprotein profiles. We also evaluated pro-inflammatory cytokines and PCSK9—a core component of the HDL proteome that is highly impacted by inflammation (Feingold et al., 2008; Burnap et al., 2021)—in the NDI-HDL characteristics relationship. We hypothesized that NDI would be associated with pro-inflammatory cytokines, PCSK9, and HDL characteristics. Additionally, we investigated associations between HDL characteristics and immune cell-based epigenetic clock biomarkers including GrimAge and PhenoAge, which have been previously reported as predictors of CVD and all-cause mortality (Levine et al., 2018; Lu et al., 2019; Joyce et al., 2021; Ma et al., 2022). We hypothesized that NDI-related HDL characteristics would be associated with these epigenetic markers, providing one of the first studies to examine associations between neighborhood socioeconomic deprivation, HDL, and epigenetic biomarkers of aging.
