High molecular weight genomic DNA is isolated from specific cell types from the mouse brain. The INTACT protocol is used to isolate nuclei from astrocytes, neurons, or microglia, and the Qiagen HMW MagAttract kit is used to isolate gDNA from the nuclei. HMW DNA is sequenced on the Oxford Nanopore PromethION platform using Kit 14 sequencing chemistry and R10.4.1 flow cells.
