The mads-box mutant line showed an abscisic acid (ABA)-insensitive phenotype. The AGAMOUS-LIKE 67 (AT1G77950) gene encodes a transcription factor that is nuclear-localized, as observed by transiently transformed epidermal onion cells, specifically expressed in seeds and involved in an ABA signaling pathway during seed germination (Fernández-Arbaizar et al. 2012). SALK_050367 seeds were obtained from the Arabidopsis Biological Resource Center, ABRC. To analyze AGAMOUS-LIKE 67 regulation of gene expression in a developmental time window of ABA responsiveness following breakage of seed dormancy (Lopez Molina et al. 2001, 2002), wild-type Col-0 and mads-box mutant seeds were stratified in water (4ºC, dark) during 3 days, and then transferred to light (22ºC, percival) in the presence of 5μM ABA for 3 hours. Three biological replicates were performed per seed type in order to get consistent results.
