BARseq using the pooled prototrophic single-gene deletion library was performed in triplicate in continuous culture for 240 hours and samples were collected every 24 hours for a total of ten timepoints per replicate per condition. Five different conditions were tested: 1) Ammonium-Sulfate limitation, 2) Glucose limitation, 3) Switching between Ammonium-Sulfate limitation and Glucose limitation every 30 hours, 4) Ammonium-Sulfate limitation with 40uM Ammonium-Sulfate pulse every three hours, and 5) Ammonium-Sulfate limitation with 40uM Glutamine pulse every three hours.
