A chromosome substitution line, CS-B25, was generated by replacing chromosome pairs 25 of G. hirsutum inbred TM-1 with a pair of chromosomes 25 from G. barbadense double haploid 3-79 lines. CS-B25 had been shown to have positive traits for fiber quality (longer, stronger and finer fiber) than the recurrent parent TM-1. In this project, a functional genomic approach will be utilized for cotton fiber improvement by using the chromosome substitution line CS-B25. The genes differentially expressed in 10-DPA fibers of CS-B25 will be identified by using a powerful molecular tool- high-throughput RNA sequencing. The expression of these genes in fibers will be further validated by qRT-PCR assays. These identified genes and KEGG (Kyoto Encyclopedia of Genes and Genomes) pathways will reveal the mechanisms of fiber development during the elongation stage and offer novel genes as useful tools for fiber quality improvement.
