Drug induced genome instability

Drug induced DNA damage in normal cells can activate error prone DNA repair pathway which leads to mutations and large-scale chromosomal rearrangements. In this study we used both Duplex Sequencing and DiGIT assay to evaluate mutations and large-scale genome instability, respectively in bone marrow and duodenum cells extracted from Sprague-Dawley rats treated with several different drugs. Our studies show that the combination of these two NGS-based approaches allows direct assessment of drug-induced genome instability and mutagenesis, the two hallmarks of human cancer.
