Whole genome sequencing of bacterial strains from Lake Zurich

Bacterial strains were isolated from surface waters from Lake Zurich in Spring 2023 (15-05-2023) in agar plates amended with modified artificial lake water (ALW, Le Moigne et al., 2023) supplemented with cellobiose (1mM final concentration). Briefly, 1mL of lake water was inoculated onto the agar plate and incubated at 18C in dark conditions for 3 weeks. The resulting colonies were transferred to a new agar plate using the standard T-streak method three times. Isolated colonies were then cultured in ALW using glucose as a sole carbon source (100 uM final concentration) and cryopreserved as described elsewhere (Rain-Franco et al., 2021).
