In this study, a mutant strain (ZMP) with a complete PPP was constructed by engineering Tkt and Tal into ZM4. A pEZ plasmid containing a xylose isomerase (XI) gene with improved xylose utilization ability was electroporated into ZMP. The resultant ZMPS strain was then adapted in xylose medium by adaptive laboratory evolution, with a mutant strain S38 obtained which can utilize xylose efficiently. RNA-Seq based transcriptome was carried out for S38 and an intermediate strain S8 to explore the mechanism of efficient xylose utilization.
