To address multidrug-resistant <i>S. zooepidemicus</i> mastitis, we engineered ClyU, a chimeric lysin fusing the Ply30 catalytic and Ply1228 binding domains. ClyU exhibited robust stability and superior bactericidal activity against <i>S. zooepidemicus</i>, <i>S. equi</i>, and <i>S. suis</i> <i>in vitro</i>. In a murine model, ClyU treatment significantly reduced bacterial burdens and inflammation without eliciting toxicity or resistance. These findings position ClyU as a potent, stable, and safe therapeutic candidate for the dairy industry.
