Transcriptome analysis of mouse lung adenocarcinomas with K14 low or high expression

Metastasis of lung cancer is the leading cause of death. Elucidate how the metastases happen and progress can potentially provide us avenues to prevent and inhibit metastasis in clinical treatments. Here we treated KrasG12D/Trp53L/L (KP) lung cancer mouse model with lysyl oxidase inhibitor which could induce extracellular matrix deprivation and tumor cells death. Interestingly, increasing amount of K14 positive cells in adenocarcinomas were observed, accompanied with several metastases emerging. Next in vitro and in vivo experiments demonstrated K14 high cells were more easily to metastasize than K14 low cells. We dissected several KP lung tumors and then checked the expression of K14 by immunostaining and real time PCR. Based on the K14 levels, we picked out three K14 low expression tumor samples, and three K14 high expression samples. Total RNA was isolated from tumors using Trizol reagent (Invitrogen). The sequencing was performed on Illumina Hiseq2500 V4 platform.
