To further verify whether the drug-resistant plasmid in the wild-type strain 20K-383 is a conjugative plasmid, YZ6 strain was used as the recipient strain, and the corresponding conjugants were screened using tigecycline.In conjugant TC383-1, both tet(A)v and blaNDM-1 are co-localized on a plasmid approximately 162,681 bp in size (designated as pTC383-1). As shown in Figure 5-3, the plasmid backbone sequence of pTC383-1 is derived from the original recipient strain YZ6's plasmid pYZ6_P1, which is approximately 122 Kb in size. Additionally, it contains a multidrug resistance gene cluster approximately 40 Kb in size. It can be observed that both tet(A)v and blaNDM-1 are co-localized within this 40 Kb multidrug resistance gene cluster.
