Multiplexed sequencing of yeast yASS1 expression cassette. yASS1 encodes the human ASS protein, using a codon-optimized derivative of the human ASS1 cDNA sequence. Derivatives of yASS1 were introduced into haploid yeast strains so that each encoded a version of human ASS with one amino acid changed. Multiplexed sequencing using the MinION platform was used to determine the altered codon, and associated amino acid change, in each yeast transformant
