To test the role of osteocytic MMP13 in bone health, we generated mice withosteocyte-specific ablation of MMP13. Homozygous MMP13fl/fl mice have loxP sites flanking exons 3, 4, and 5, which encode the enzyme’s active site. Hemizygous DMP1-Cre+\- mice express Cre predominantly in osteocytes. Mice were bred to generate wildtype (WT, DMP1-Cre-/-; MMP13fl/fl) and MMP13ocyKO (DMP1-Cre+/-; MMP13fl/fl) mice. For this study, male mice humerus were utilized. Osteocytes were sequenced from WT and MMP13ocyKO mice (n=4/group; 8weeks).
