Using our previously established homozygous mutant mouse ESC bank, we identified a previously uncharacterized novel gene AU019823, which we named “attenuated hematopoietic development (Ahed)”, whose null mutant ESCs exhibited attenuated production (70-fold fewer) of blood cells during the hematopoietic differentiation supported by coculture with OP9 stromal cells. Both mutant and revertant (almost identical to wild-type) ESCs normally differentiate into Flk-1-positive early mesodermal cells in vitro. In this study, to explore the function of this gene, we analyzed the gene expression profile of Flk-1-positive cells with these two genotypes. Overall design: In this study presented here, a total of two samples were analyzed: Flk-1-positive cells differentiated from AU019823 gene-trapped (biallelically disrupted) ESCs and their revertant (almost identical to wild-type) ESCs were subjected to gene expression analyses.
