资源简介:
Myeloid-derived suppressor cells (MDSC), a heterogeneous population of myeloid cells, accumulate in the melanoma microenvironment. With the ability to inhibit anti-tumor T cell responses, MDSC have been shown to promote immunosuppression, enhancing tumor progression and tumor cell resistance to the immunotherapy. Novel markers are still needed to define MDSC. In this study,we wanted to find possible markers for M-MDSC by comparing cd14+ monocytes from melanoma patients and healthy donors by scRNA-seq. Overall design: Peripheral blood mononuclear cells (PBMC) were isolated from whole blood of 2 treatment-naive stage IV melanoma patients and 2 healthy individuals via density gradient centrifugation.CD14+ monocytes were isolated from PBMC by MACS (Miltenyi Biotec) according to the manufacturer's instructions. 10x Genomics Chromium single-cell 3' library kit was used to prepare sequencing library. 10000 cells were loaded on the chips.
