The predictive value of CD14 gene promoter region 159C / T polymorphism combined with red and white blood cell ratio for RA disease activity

Objective To investigate the application value of CD14 gene promoter region 159C / T polymorphism and blood routine red and white line ratio in the diagnosis of rheumatoid arthritis ( RA ) and the evaluation of disease activity.Methods From February 2022 to October 2023,203 patients with RA ( RA group ) and 192 healthy people ( control group ) were selected from the Department of Rheumatology, Gansu Provincial Hospital of Traditional Chinese Medicine, Qingyang People 's Hospital and Lanzhou University Second Hospital.The promoter region -159C / T polymorphism of CD14 gene was detected by PCR - RFLP.The DAS28 score was used to evaluate the severity of RA patients ( mild, moderate, and high activity groups ).The blood cell derived parameters RLR, HLR, RNR, HNR, RWR, RMR and HMR were calculated,and collected the clinical data and immunological indicators,and to all the data for statistical analysis.Results RLR and HLR in group RA were higher than those in control group (P<0.05), while RNR, HNR, RWR, RMR and HMR were lower than those in control group (P<0.05).The difference of RWR in RA mild, moderate and high activity groups was statistically significant (P<0.05), which was negatively correlated with disease activity.There was no significant difference in the distribution of -159C/T genotype in the promoter region of CD14 gene between RA group and control group (P>0.05), but there was significant difference in the frequency of C and T genes in different RWR values,Χ2=7.907,P<0.05, and the frequency of C gene increased gradually with the decrease of RWR value.Conclusion CD14 gene promoter region-159C / T gene frequency combined with RWR may be a new index to predict RA activity, and RWR is negatively correlated with RA disease activity.
