16S rRNA V3-V4 amplicon sequencing of Holstein dairy cows rumen microbiota

The effect of water-soluble cellulose acetate (WSCA) on Holstein dairy cows rumen microbiota was evaluated by the rumen simulation technique (RUSITEC). Rumen fluid and solid contents were collected from 2 ruminally fistulated Holstein cows. The rumen inoculum was a mixture of rumen fluid and artificial saliva at a 8:5 ratio. Continuous culture with RUSITEC was performed using 650 ml of rumen inoculum, 70 g of rumen solid contents and 12 g of formula feed. After 120 h of acclimation period, 5.1 g of cellulose or 2.55 g of cellulose and 2.55 g of WSCA were added to formula feed individually, as control group or treatment group. Samples were collected 24 h after start of the test period and used for microbial analysis. Culture was performed in quadruplicate.
