<b>Background/Objectives:</b> Impaired DNA repair has been linked to Parkinson’s disease (PD), especially under oxidative stress and environmental exposures. This study evaluated five polymorphisms in nucleotide excision repair (NER) pathway genes and their association with PD in a northern Mexican mestizo population.<b>Methods:</b> We performed a case–control study with 137 PD patients and 137 matched controls. DNA was extracted from blood, and genotyping of ERCC1 (rs11615), ERCC2 (rs13181), XPA (rs1800975), XPC (rs2228001), and XPF (rs1799801) was conducted using TaqMan assays. Logistic regression adjusted for age, sex, and pesticide exposure was used to test associations.<b>Results:</b> Pesticide exposure was more frequent in PD patients (OR 2.08, 95% CI 1.18–3.68; p = 0.01). The XPC rs2228001 C/C genotype was associated with higher PD risk in males (OR 4.23, 95% CI 1.33–13.49; p = 0.015), independent of uric acid and pesticide exposure. Other polymorphisms showed no significant associations. Male patients also had lower uric acid levels (p = 0.047).<b>Conclusions:</b> The XPC rs2228001 variant may contribute to male-specific PD susceptibility, particularly with pesticide exposure. These findings highlight the role of DNA repair pathways and the interaction of genetic and environmental risk factors in PD.
