Sequencing of histone H4-HaloTag incorporated genomic regions.. Homo sapiens

Recent advanced live-cell imaging techniques have revealed the dynamic behavior of chromatin in living cells. Especially, our single-nucleosome imaging studies have revealed the genome-wide local chromatin motion and its heterogeneity, which can provide a higher-order regulation of genome functions. Individual nucleosomes in living cells are commonly labeled by tagged core histones (e.g., histone H4 tagged with HaloTag). To determine the genomic localization of the nucleosomes labeled with H4-HaloTag in HeLa cells, we pull-downed the H4-HaloTag nucleosomes and sequenced their genomic DNAs.
