Grapevine downy mildew (DM) is a destructive oomycete disease in viticulture worldwide. MrRpv1 is a typical TIR-NBS-LRR type DM disease resistance gene cloned from Muscadiana rotundfolia. But the molecular basis of DM resistance mediated by MrRpv1 remains poorly understood. MrRpv1-transgenic plants were developed with DM resistance in the genetic background of DM-susceptible Vitis vinifera cv. Shiraz. Comparative transcriptome analysis was used to compare the transcriptome profiles of two different genotypes after DM infection. The transcriptome response during P. viticola infection is faster and more genes are induced in MrRpv1-transgenic Shiraz than in wild type plants. In DM-infected transgenic plants, burst of Ca2+ and ROS is the earliest response. Function analysis of differentially expressed genes revealed that key genes related to multiple phytohormone signaling pathways and secondary Metabolisms were activated during infection. Co-expression network and motif enrichment analysis showed that WRKY and MYB transcription factors strongly coexpress with STS genes. Taken together, these findings indicate that multiple pathways play important role in MrRpv1-mediated Downy Mildew defense.
