Ralstonia solanacearum strain ES5-1 genome sequencing

Here we report that a highly virulence strain named ES5-1 was isolated from tomato bacterial wilt tissues, which could break the resistance of disease resistant tomato cultivar Hawaii 7996. We found ES5-1 strain belongs to phylotypes III sequence suqver 1 and RipV2, one of the type III effectors existed specifically in R. solanacearum phylotypes II strains, interacts with tomato helper NLR N REQUIREMENT GENE 1 (NRG1) to overcome plant resistance. RipV2 encodes a novel E3 ubiquitin ligase and targets tomato NRG1/EDS1/SAG101 module for degradation via Ubiquitin/26S proteasome pathway. Expression of RipV2 in Hawaii 7996 roots dampen plant immune responses and bacterial wilt defense. RipV2 was required for ES5-1 full virulence on Hawaii 7996 but not SlNRG1-silenced tomato, suggesting SlNRG1 are the bona fide target of RipV2 in plants.
