To further understanding of miRNA expression during radioprotectation, we used microarray analysis to determine the differential expression of miRNAs in the bone marrow of mice exposed to high-dose IR with and without pretreatment with three types of radioprotectants: Amifostine (WR2721), CBLB502, and Nilestriol, as well in the hypothalamus of mice exposed to IR with and without pretreatment with amifostine. Overall, amifostine treatment led to a similar expression pattern of miRNAs to that of CBLB502 but not to that of nilestriol in bone marrow. The radioprotectants exhibited mostly opposite effects against IR-induced miRNAs. These results expand the biological function of miRNAs in mediating the activity of radioprotectants, further offering insights into the molecular mechanism underlying radioprotection. Radiation induced miRNA expression in mice bone marrow and hypothalamus pretreated with amifostime, CBLB502 and nilestriol was measured at 12 hours after exposure to doses of 8 Gy gamma-rays. The bone marrow from three animals were pooled in one sample for each treatment.
