We used CRISPR/Cas9 to knock in three human metabolic disease risk genes (PNPLA3I148M-GIPRdn-hIAPP) into the H11 locus of the Bama pig genome. Combined with high-fat and high-sucrose diet (HFHSD) induction, we aimed to construct a large animal model for metabolic disease, and further use it as a material to study the molecular mechanism of metabolic disease-induced cardiac injury. The triple-transgenic pigs (TG) displayed lipid metabolism disturbance, myocardial injury including tissue edema, myofiber rupture and mitochondrial damage after a long-term HFHSD intervention. This RNAseq study compared the 3 TG with 3 wild-type controls (WT) that were also induced by HFHSD but had no pathological changes, providing gene expression data for studying the molecular mechanisms of metabolic disease-induced cardiac injury.
