LFMT profiling

We recently published a multicenter, randomized, double-blinded, non-inferiority trial aiming to compare the efficacy of lyophilized microbiota transplantation versu lyophilized faecal filtrate to prevent recurrent clostridioides difficile. This project utilized human peripheral blood mononuclear cells (PBMCs) collected from the trial.Briefly, approximately 10,000 human PBMCs from two recurrent Clostridioides difficile infection patients at baseline and 8 weeks post-lyophilized fecal microbiota transplantation were used for single-cell RNA sequencing. Cells were fixed using the Evercode Fixation V3 kit (Parse Biosciences) according to the user manual. Then, two sublibraries were generated using the Evercode WT Mini kit (Parse Biosciences) and sequenced on an Illumina NovaSeq X Plus sequencer with 150-base pair (bp) paired-end reads, yielding approximately 100,000 reads/cell.
