The aim of present study was to compare the in vitro antioxidant and antibacterial properties of carboxymethyl cellulose (CMC) films containing resveratrol (RES) and eugenol (EUG), alone and in combination, and to calculate the interactions between them. At first, the total phenolic content of CMC films were evaluated. Then, their antioxidant and antibacterial effects of films were determined using DPPH, reducing power, disc diffusion and broth dilution methods. Finally, concentrations of RES and EUG which showed better results in the CMC films were added in combination forms to calculate their antioxidant and antibacterial interactions. The results showed that addition of RES and/or EUG to CMC films increased the total phenolic content, free radicals scavenging activity, reducing power and antibacterial activities of the films (P≤0.05). Gram-positive bacteria were more susceptible than Gram-negatives. In addition, the combined use of RES and EUG in CMC films had synergistic antioxidant and antagonistic antibacterial effects. The best results belonged to the film containing RES (8 µg/mL)+EUG (8 mg/mL) (P≤0.05). Considering the results of the present research, we can utilize CMC biodegradable film containing RES and EUG as a natural active packaging in food industry.
本研究旨在对比单独或复配添加白藜芦醇(resveratrol, RES)与丁香酚(eugenol, EUG)的羧甲基纤维素(carboxymethyl cellulose, CMC)薄膜的体外抗氧化与抗菌性能,并解析二者间的相互作用。首先,本研究测定了各CMC薄膜的总酚含量;随后采用DPPH法、还原力法、纸片扩散法与肉汤稀释法,分别评估薄膜的抗氧化与抗菌活性;最后选取在CMC薄膜中表现最优的RES与EUG浓度进行复配,以计算二者的抗氧化与抗菌相互作用。结果显示,向CMC薄膜中单独或复配添加RES与EUG,均可显著提升薄膜的总酚含量、自由基清除活性、还原力与抗菌活性(P≤0.05)。革兰氏阳性菌较革兰氏阴性菌对该薄膜更为敏感。此外,CMC薄膜中RES与EUG复配使用时,可产生协同抗氧化效应与拮抗抗菌效应。其中,添加8 µg/mL RES+8 mg/mL EUG的薄膜性能最优(P≤0.05)。综合本研究结果,含RES与EUG的CMC可降解薄膜可作为天然活性包装材料应用于食品工业。
