A two-step co-transformation strategy was performed to identify the critical component in limiting conjugation efficiency. A chuB::erm marker was introduced into high frequency conjugation strain C. jejuni CG8486 as the co-transformation selection marker. The chuB::erm chromosomal DNA was used to transform C. jejuni NCTC 11168, which displays extreme low conjugation efficiency. Transformants are expected to carry other donor (CG8486) DNA fragments, including HFC factor. Potential high frequency conjugation transformants were sequenced by MiSeq.
