Using natural isolates of Saccharomyces cerevisiae, we sequenced an advanced intercross population under selection in order to increase genetic mapping resolution. We created an advanced intercross population starting from an oak/vineyard F1 using 11 rounds of meiosis and random mating. Allele frequencies and linkage in the advanced intercross population were determined by sequencing the final (F12) population and isolated clones. To enrich for QTL conferring increased growth rate, we maintained the advanced intercross population in replicate glucose-limited chemostats at a low (D=0.18 hr-1) or high (D=0.35 hr-1) dilution rate. A lower dilution rate results in a lower steady-state glucose concentration in the chemostat. We also pooled a panel of F2 segregants and grew them in the same chemostat conditions. We collected and sequenced multiple samples from each chemostat over 20–40 generations.
