Gene expression profile of purified peripheral CD14+CD163+ macrophages was analyzed via RNA-Seq in the presence or absence of inflammatory stress to assess the impact of TET2 disruption on gene expression in macaque hematopoiesis. Mobilized CD34+ cells were treated with CRISPR/Cas9, followed by transplanted autologously to generate a non-human primate model of clonal hematopoiesis. Peripheral CD14+CD163+ macrophages were purified at early (4.5 months) and late (19 months) timepoints post-transplantation and incubated for 8 hours in the presence or absence of inflammatory stimulators, and then RNA was extracted and analyzed by high-throughput sequencing.
