After treatment with hydrogen peroxide, the transcription levels of each gene in Mtb ∆ orn ∆ CRISPR double knockout strains were detected in response to CRISPR system or Cas10 HD domain mutants. After treatment with 10 mM H ₂ O ₂ for 4 hours, Mtb cells were collected at OD ₆₀₀=0.4 and immediately frozen and ground in liquid nitrogen. Total RNA was extracted using TRIzol reagent (Invitrogen) according to the manufacturer's instructions. Subsequently, Ribo clean rRNA Deletion Kit Mega (Vazyme) was used to remove ribosomal RNA. The chain specific RNA library was constructed using the VAHTS Universal V8 RNA seq Library Prep Kit for Illumina (Vazyme) and sequenced on the Illumina NovaSeq 6000 platform, obtaining a 150 bp double ended read length.
