Oxymatrine (OMT), one of the major tetracycloquinolizindine alkaloids extracted from Sophora roots, possesses antitumor and anti-inflammatory activities. OMT inhibits the AP endonuclease activity of APE1, thereby leading to the accumulation of DNA damage that results in the inhibition of proliferation and induction of apoptosis in A549 cells. However, the mechanism by which OMT influences the transcriptional function of APE1 remains unclear. Therefore, in this study, we employed cutting-edge molecular biology tools and next-generation RNA-sequencing technology to investigate the antitumor mechanisms of oxymatrine.
